Title: Untargeted or targeted, what is the best approach in metabolomics

Abstract: Untargeted and targeted metabolomics are two complementary approaches, which when combined can achieve in-depth and comprehensive lipidome characterisation and quantification. The two approaches are usually performed on two different MS platforms, triple-quadrupole (QqQ) type MS and high-resolution MS. Here, we discuss a development of a robust workflow merging both targeted and untargeted approaches on a single methodology using liquid chromatography coupled to quadrupole-time of flight system (LC-QqTOF) platform with scheduled parallel reaction monitoring (sPRM). sPRM assays integrate both untargeted profiling from MS1 scan and targeted quantification obtained from MS/MS data. With scheduled algorithm and maximum 100 precursors multiplexed per duty cycle, sPRM assay can achieved a high-throughput targeted monitoring of more than 250 lipid species within a single injection. This workflow has applied to complex lipidomics study, which enabled the identification and quantification of more than 500 lipid species from 25 lipid classes in a barley root extract.

Bio: Thusi is an analytical chemist, currently working as a  Field Application Specialist with SCIEX in Oceania region. While working in University of Melbourne, she has developed analytical methodologies based on LC-MS for metabolomics and lipidomics. Thusi played an instrumental role in establishing Australian Lipid Meeting (ALM) along with leaders in lipidomics community.  She has also been involved in ANZMN and  ASPL and contributed to organizing a number of conferences in metabolomics and lipidomics. She is also an active member in promoting women in science, Females in Mass Spectrometry (FeMS), as well as diversity and inclusion events.
